If you want to associate ed2k with amule in firefox, install amule-utils first.

Enter about: config in the address bar of firefox

Create a new Boolean named network. protocol-handler.external.ed2k with a value of true
Create a new String named network. protocol-handler.app.ed2k with a value of/usr/bin/ed2k. amule or ed2k. amule or ed2k (My amule 2.1.3 is ed2k)
