I. preparation of sequence text
Before constructing the tree, the target gene sequence is saved separately into a txt text file (or all the sequences are saved in the same txt text, the "> Gene name" can be used as the first line, and then the sequence of the edited gene is repeated ), and the series contains only Sequence letters (ATCG or amino acid shorthand letters). The filename name can be edited by your thoughts.
Second, serial import to Mega 5 software
(1) Open the Mega 5 software, the interface is as follows

Select the analysis sequence type (if it is a DNA sequence, click on DNA, if it is a protein sequence, click Protein)
A new dialog box appears, creating a new data file
Select Sequence Type
Import sequence
The import sequence succeeded.
(3) Sequence alignment analysis
Click the "W" tool in the toolbar to perform a comparison analysis, and delete both ends after the end can not be completely aligned to the base
(4) System development Analysis
Close window, choose Save file path, custom file name
Three, System development tree construction
According to different analysis purposes, choose the corresponding analysis algorithm, this example takes the N-J algorithm as an example
Bootstrap Select 1000, click Compute, start counting
When the calculation is complete, the system development tree is generated.
