"Nature Biotechnology" mainly published in the field of biotechnology, the original innovative research results, 2015 JCR Impact factor of 41.514, has surpassed the main issue of "Nature" (41.456). The study found a wider range of gene editing tools for genome site editing. This tool is completely different from the RNA-guided CRISPR/CAS9 gene editing technique: This argonaute (abbreviated Ngago) from the source of the ancient bacterium, using short-chain DNA as a guide, really enables the genome to be cut anywhere, and the possibility of gene editing is pushed into a broader position. This technology has the following clear advantages: 1. Wizards are designed to be simple: the short chain single-strand DNA wizard can be synthesized like a synthetic PCR primer; The wizard can directly transfection cells and tissues without building a wizard expression vector. 2. Any location within the editable genome: the target selection of the Cas9 genome is limited by the PAM region and the GC-rich area. The Ngago has no restriction on the target selection, and can effectively introduce double-stranded fracture in any position of the genome. 3, since the guided nucleic acid is DNA rather than RNA, it avoids the failure or off-target effect of RNA that is prone to complex two-level structures. 4, it has higher cutting efficiency to the DNA which is free from the nucleus. The result is China's first "China-created" cutting-edge biotechnology, breaking the patent monopoly of foreign gene editing technology, the level of research can be comparable to international first-class universities in the same field, such as the University of California, Berkeley, Jennifer a doudn and MIT Zhangfeng Professor's work. The technology can be used for the accurate gene transformation of microorganisms, plants and animals, and the "gene therapy" of hepatitis B, AIDS or some hereditary diseases, which is of great significance in the editing and rebuilding of human blood and organs, and has important application value in the fields of medicine, agriculture, animal husbandry and other industries.

Here we report this the natronobacterium gregoryi argonaute (NgAgo) is a dna-guided endonuclease Suita ble for genome editing in human cells. NgAgo binds 5′phosphorylated single-stranded Guide DNA (GDNA) of ~24 nucleotides, efficiently creates site-specific DNA D Ouble-strand breaks when loaded with the GDNA. The Ngago–gdna system does not require a protospacer-adjacent motif (PAM), as does Cas9, and preliminary characterization suggests a low tolerance to guide–target mismatches and high efficiency in editing (G+C)-rich genomic targets.
